Concept About Indomethacin
Pharmacological actions
A. Potent anti-inflammatory agent than aspirin
B. Anti-pyretic action and
C. Analgesic activity.
- Rapid absorption form the GIT.
- No effects on uric acid excretion (safe in patient with gout)
- Does not modify the action of oral anticoagulants.
- It antagonises the antihypertensive action of diuretics, beta-blockers. ACE-inhibitors.
Indications of indomethacin
1. Acute Gout
2. Musculoskeletal disorder
3. Rheumatoid arthritis
4. Ankylosing spondylitis
5. Osteoarthritis
6. Patent ductus arteriosus in neonates
7. Pericarditis
8. Bartter’s syndrome.
9. Pleurisy.
Dose: 25–50 mg 2-3/day
Adverse effects of indomethacin
a. GIT: Peptic ulcer, Nausea, Vomiting
b. CNS: Severe frontal headache, Dizziness, Vertigo, Mental confusion
c. Haematological: Neutropenia, Thrombocytopenia, Thrombocytopenia
d. Eye: Retinopathy/ank
e. Hypersensitivity; Rash, Itching

Contraindications of indomethacin
1. Pre-existing gastric ulcer.
2. Pregnancy. nursing mother.
3. Children.
4. Renal disorders.
5. Psychiatric disorders
6. Epilepsy
7. Parkinsonism
Ibuprofen
Pharmacological actions of ibuprofen
1. Potent analgesics than aspirin or paracetamol.
2. Anti-inflammatory action.
3. Anti-platelet action.
Mechanism of action:
Potent inhibitors of cyclooxygenase.
Indication of ibuprofen
1. Rheumatoid arthritis.
2. Osteoarthritis.
- Adverse effects and contraindication are same as aspirin
- Advantage over aspirin: less incidence of GIT ulceration with low dose.
