Concept About Pethidine
1. Pethidine is also known as meperidine.
2. It is a synthetic substitute for morphine.
3. Less potent analgesic than morphine.
4. Absorbed from Gastrointestinal tract (GIT).
5. Plasma protein binding capacity: Partially.
6. Bioavailability: 40-60% with oral route but more in parenteral route.
7. Short duration of action (2-3 hours); hence pethidine is preferred to morphine for analgesia during labor.
8. Metabolized in liver by hydrolysis to pethidine acid or demethylation to norpethedine & hydrolysis to norpetherdinie acid, followed by conjugation with glucoronic acid.

- Mydriasis
- Antispasmodic action
- Atropine like side effects
- Anti-histaminic action.

11. Uses:
Used as analgesic in –
1. Severe pain
2. Renal colic
3. Minor surgery
4. Ophthalmology, dentistry
5. Also used in-
6. Preoperative medication and
7. As an adjunct to anaesthesia
8. To relieve pain of labour
9. Spasm of GIT, uterus & urinary bladder.
Pethidine does not suppress cough usefully
