Drug Dose

Amount of drug or medicinal preparation to be administered at one time is called dose. It is the amount of the drug. Which is required to produce certain measurable biological response, either at once or after sometime of administration.

Classification of Dose:

➤ Therapeutic dose: Dose required producing the optimal therapeutic effect.

➤ Maximum dose: Largest dose of drugs that is safe to administration and produce no toxic effect.

➤ Minimum dose: Smallest dose of drug that will be effective.

➤ Booster dose: An additional dose to increase the protection afforded by the original series of injection.

➤ Loading dose: The loading dose is one or a series of doses that may be given at the onset of therapy.

➤ Maintenance dose: Dose required to maintaining the desired effect that is achieved by preceding dose.

  • Maintenance dose – Dosing rate x Dosing interval
  • (Maintenance dose is governed by CL and t½ of drug)

➤ Test dose: Amount of drug given initially to see the response of drug to tissue. It is given in

  • To observe the sensitivity of tissue to the drug (example: antibiotic hypersensitivity)
  • To see the potency and nature of response of a drug

➤ Fatal dose: Amount of dose that causes death of 100% of experimental animals.

➤ Lethal dose: Amount of dose that causes death of some of experimental animals.

➤Toxic dose: Dose that causes signs and symptoms of drug toxicity.

➤ TD50 (median toxic dose): The dose required to produce a particular toxic effect in 50% of test animals is called the TD50
